A model for tumor cells cocultivation with human dermal fibroblasts (two-dimensional multicellular spheroids) was established. Using this model, it is shown that fibroblasts direct the movement of tumor cells and contribute to their polarization for further migration, also detected colocalization the components of mTOR signaling cascade with intermediate filaments. the monoclonal antibodies against the protein component Rictor mTORS2 complex was sharacterized. Using these antibodies the localization mTORS2 complex in association with the cytoskeleton (cytokeratins) circumferentially cells membrane space was shown. The existence of a new kinase isoforms of ribosomal protein S6 - S6K1 / d15 in humans and mice was proved, and it was shown that their expression is enhanced in stress conditions (cell growth in a poor nutrient medium and growth factors).
