Aim - to determine the effect of endogenous and exogenous estrogens on biochemical and functional disorders in the cardiovascular system of female rats with diabetes. Methods: pathophysiological, biochemical, spectrophotometric, pharmacological, immunological, organic synthesis, physical and chemical analysis.It was established that estrogen deficiency increases the negative effect of diet on the development of insulin resistance, but does not affect the reducing of function pancreatic beta-cells induced by streptozotocin. Development and severity of both dyslipidemia and visceral obesity were not dependent on the level of endogenous estrogen in female rats with experimental type 2 diabetes. Estrogen deficiency enhances the disturbance of the oxidation-reduction balance in blood serum and mitochondria of cardiomyocytes in female rats with type 2 diabetes. It was established that activity of NO-synthase and hemeoxygenase significantly reduced in homogenate vessels compared with intact control independently of the level of endogenous estrogen in rats with type 2 diabetes. Estrogen deficiency increases the imbalance of hemostasis coagulation due to rising prothrombotic factors in diabetic rats. Estrogen deficiency aggravates functional disturbances in the cardiovascular system of female rats with type 2 diabetes. Type 2 diabetes eliminates the protective effect of exogenous estrogen on glucose homeostasis disorders, visceral obesity and oxidative stress in the vascular endothelium of animals with estrogen deficiency. The detected properties of selective estrogen receptor modulators compounds PE0607 to inhibit lipid metabolism, immunological and hemocoagulation dysfunction, to normalize the activity of nitric oxide nitrogen in the blood vessels, to prevent the development of mitochondrial dysfunction and oxidative stress in the heart, to restore heart rhythm and intraventricular conductivity, indicate the perspectives of its use as an additional gender-specific agent for the prevention and treatment of diabetic cardiovascular complications in women.
