The aim of the study: To investigate the role of the peripheral and central dopaminergic system in the mechanisms of the alterations of the intestinal barrier function in IBD. Меthods: chemical destruction of the peripheral and central dopaminergic neurons, modelling of the experimental ulcer colitis, pharmacologic studies, the determination of the endothelial permeability in vivo, mieloperoxidase activation, proteins content by Western Blot analysis. It was studied that both in rats with experimental colitis and chronic ulcer colitis patients was inhibited dopamine synthesis (down-regulated level of TH in colonocytes) and was changed DAT, D2 and D3 receptors expression in colonic mucosa. It was shown for the first time, that destruction of the central peripheral dopaminergic neurons led to aggravation of the experimantal colitis flow. The level of the aggravation of the experimental colitis proportional to the level of the central dopaminergic system destruction. The opposite result was received under destruction of the peripheral dopamniergic neurons. One of the mechanisms of the protective action of the dopaminergic system in the pathogenesis of the ulcer colitis is supporting of the intestinal barrier function. The protection of the endothelial barrier is partially mediated by activation of the central D2 dopaminergic receptors and by inhibition of the adhesive potential of the endothelial cells through the ICAM-1 expression. It was prooved that usage of the preparation with dopaminergic activity (agonists of the D2 and D3 receptors), is efective in heeling of the ulcers in experimental colitis and could be recommended for clinicla studies.
