The object of research - potato leaf roll virus, mammalian cell cultures, specific antibodies. The purpose - by studing of features of the system PLRV- animal cell, studing morphological, physical, chemical, biological and antigenic properties PLRV strains, which circulating in Ukraine, improved methods of diagnosis. Research Methods - virological, immunological, serological, physicochemical and methods of biometric processing results. It is shown that productive infection PLRV at the mammalian cells culture of enables the selection of pure virus isolates from potato plants with mixed infection. Obtain pure populations of potato leaf curl virus, which replenished collection Institute. It is proved that the cultivation of a potato leaf roll virus in mammalian cell culture does not affect its pathogenic properties and can transmit the virus to the plant. It was established that the PLRV were susceptible cell cultures inoculated lines swine embryonic kidney (SNEV), Syrian hamster kidney (BHK-21), pigs testicles (TAP), kidney bull (MDVK) and rabbit kidney carcinoma (RK- 13). Research PLRV features a reproduction cycle in cell culture BHK-21. It was established that adsorption of the virus required cell receptor protein or glycoprotein nature. As a result of studies of physical and chemical properties PLRV isolates revealed that viruses resistant to media with pH values from 4.0 to 10.0 and at 50°C termorezystentni at the absence of divalent magnesium ions, TTI is in the range 60-65°C. The optimum temperature for PLRV reproduction is 24°C and above. It was demonstrated that virus isolates twisting leaves of potato isolated from various sources (aphids, leaves and tubers) 100% related to each other. It was established that the use of neutralization in mammalian cell culture, can quickly and reliably diagnose PLRV in plant material and aphids.
