Objective: To study the level of methylation eEF1A in carcinogenesis. To create eEF1A point mutants where lysine residues at 55, 79 and 165 positions are replaced with arginine. To start the identification of protein-partners of eEF1A, the interaction to which is dependent on eEF1A methylation. Methods of research: Chromatographic separation of proteins, mass-spectrometry, point mutagenesis based on the polymerase chain reaction. eEF1A were isolated of Lewis carcinoma, Walker carcinosarcoma and of the liver of rats. The level of methylation of lysine residues eEF1A was estimated with mass spectrometry. A tendency to a decrease in the methylation level of lysine residues eEF1A at 79, 165 and 318 positions, while the methylation level of lysine at position 55 was not changed. The results indicate the involvement of eEF1A methylation in the regulation or deregulation of cellular processes in carcinogenesis. That suggests the need for further study of its functional role. The point mutants in which the lysine residues 55, 79 and 165 were substituted with arginine were obtained. These mutants are necessary for further study of the functional significance of methylation. The proteins were identified, the interaction to which were influenced by methylation of eEF1A at 55 position, namely eEF1A1 (dimer), the other elongation factors, ribosomal proteins, chaperone HSP70, cytoskeletal proteins actin and tubulin, aminoacyl-tRNA synthetases, RNA helicase, proteins that involved in RNA processing, repair and maintenance of the structure of DNA, apoptotic proteins and others.
