During the reporting period was to study the effect of human mesenchymal stem cells (MSCh), T-lymphocytes, mesenchymal stem mouse bone marrow cells (MSCm BM) and interferon alfa-2b for the development of the tumor population adenocarcinoma cells of breast gland in monolayer and spheroid growth. Research Project: mesenchymal stem human bone marrow cells (MSCh BM) and mouse (MSCm) and tumor cells MCF-7. In this work we were used imunotsitohimicheskie, flow cytometric and molecular-biological methods, biochemical and statistical analysis methods. The aim was to characterize the influence of biologically active substances of cellular proliferation, survival, receptor profile and ability to form multicellular spheroids tumor cells in vitro and for tumor in an animal model. Tasks study was to determine the kinetic parameters of human tumor cell growth in culture when incubated with cell-derived substances (interferon alpha-2b), the expression level score tumorigenic tumor cells some markers influenced MSCm BM and T-lymphocytes, interferon alpha-2b. As well as a comparative analysis of the above parameters at different stages of cultivation in monolayer and spheroid culture. Comparison was made on the level of expression of estrogen receptor, epidermal growth factor receptor, cytokeratin and E-cadherin. Results. K medium from T-lymphocytes in general has pro-oncogenic effect on a population of tumor cells MCF-7. Since stimulates proliferation, reduces the adhesion of cells increases expression of estrogen receptor and the epidermal growth factor receptor and stimulates the formation of multicellular tumor spheroids 5 times larger than the control. K medium from MSCh conversely reduces the proliferative activity of tumor cells, reduces the ability of cells to migrate into the slurry fraction reduces the expression of estrogen receptor and the epidermal growth factor receptor. And also to MSCh environment inhibits the growth of multicellular tumor spheroids cell line MCF-7 by 60% compared with the control. High doses of cytokine IFN-?-2b (104-105 IU/ml) lower survival breast adenocarcinoma cell line MCF-7. The level of expression of nuclear receptor ER? estrogenovovogo suppressed effect of high doses of IFN-?-2b (104-105 IU/ml), thus increasing the percentage of cells in the culture do not express ER?. Simultaneously, the expression of E-cadherin receptor cells increased by 38% and expression of cytokeratin clone AE1-AE3 MCF-7 cells remained at the control level. MSCh BM also affect the receptor status of the tumor cells. Thus, upon incubation with MSCh BM percentage of EGF-R+ cells increased from 70.1% (control) to 88.3%, the amount of PAN+ cells remained at 100%. According to our results the percentage of cells expressing E-cadherin, after incubation with MSCh BM decreases from 46% (control) to 13.8% and the expression of estrogen receptors of the tumor cells increased from 35.3% in the control to 42.8% in while the number of EP- cells is not significantly reduced. Introduction cytosol mouse bone marrow cells contributed experimental animals transplanted tumor growth inhibition of 17% - 31%. Injection cytosol of mouse bone marrow cells contributed to the normalization of some indicators of biochemical blood test animals. Scientific and practical significance. From a scientific point of view, we have been investigated basic mechanisms of interaction of the tumor microenvironment and cell populations. When non-contact co-cultivation messengers appear humoral signals (interleukins, cytokines, growth factors), and mediators - tumor cell receptors, which in turn trigger a cascade of proliferative, adhesion and migration. From a practical point of view, to determine the conditions of antitumor action of MSCh BM, T-lymphocytes and interferon alfa-2b in particular at the level of an animal model will allow developm effective antitumor agents based on the cytosol of human stem cells without the risk of stimulating tumor progression and cell contamination.
