Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 Established differences in the mechanisms of activation and post-translational modification of FoxO1 and IkB in normal and EBV transformed human B lymphocytes. The revealed differences in expression of mCD150 and nCD150 isoforms in cells of B-lymphocyte and glial origin. It is shown that cells in lymphoid dominating isoform expression mCD150, and glial - nCD150 isoforms. It is shown that unlike normal B-LCL, CD40 ligation on cells XLP B-LCL does not result in phosphorylation of IkB. Expression of transcription factors profile (IRF4, IRF8, PU.1, Bcl-6, MITF, SpiB and BLIMP1) in malignant B cell lines, EBV-transformed B cell lines, obtained from healthy donors and patients with X-linked immunodeficiency (XLP) was perfomed. It was found that expression level of transcription factors IRF4, IRF8, Bcl-6 and PU.1 in malignant B cells not always correspond to the stage of differentiation, which arose block maturation of these cells, indicating that the imbalance in the regulation of these transcription factors. It is shown that the expression level of IRF4 and PU.1 transcription factors was reduced in XLP B-LCL. It was established that human beta-defensin-2 (hBD-2) affects the proliferation of cancer cells, including malignant blood cell lines Namalwa and Jurkat in the model in cellulo, as well as the expression and post-translational modification of proteins associated with cell cycle regulation, such as cyclin D1 and E, c-Abl, E2F1, pRB and others. A functional homologue of hBD-2 in mice, mBD-2 shows similar activity against Lewis lung carcinoma cells in cellulo and in vivo. hBD-2 acts as a negative regulator of transcription factors required to undergo terminal differentiation stage of B-lymphocytes (IRF4 and Blimp1) in malignant cell lines of B lymphocyte origin (BJAB and Raji). For the first time found that the CD40 receptor is a positive regulator of the expression of hBD-2 in B-cell line T5-1. At the same time, BCR ligation leaded to downregulation of beta-defensin-2 mRNA level.
