Objects of research: plasma fractions II + III and III by Kohn, prothrombin complex factors, factor VIII blood coagulation, plasminogen, albumin, immunoglobulin G, Diasorb aminopropyl, Diasorb-diol (pore size 250-1500 A), hydrophobic ligands, hydrophobic silica sorbents. Purpose: To synthesize new sorbents based on macroporous silica-inorganic matrix and hydrophobic ligands suitable for use in the process of obtaining protein preparations of plasma using the previous methods of fractionation, affinity chromatography and antiviral treatment. Methods: The methods of chemical synthesis, chromatographic methods of isolation and purification of proteases, methods of fractionation of plasma (alcohol, saline, PEG-fractionation), centrifugation, affinity and ion-exchange chromatography, gel filtration, dialysis, spectrophotometry, methods for determining the activity of proteases (of chromogenic substrates, coagulation of natural substrates), ELISA, electrophoretic methods. As a result of the work received the following scientific and practical results: synthesized derivatives of hydrophobic amino and hydroxypropyl-Diasorb with pore sizes of 250, 500, 750, 1000 and 1500 A and ligands isopropyl, butyl, octyl, phenyl; investigated the physicochemical properties of synthesized sorbents that are tipical for of macroporous silica matrix (toughness, high chemical and physical stability, lightness of regeneration, resistance to microorganisms, the possibility of sterilization by autoclaving); experiment investigated sorption-desorption properties of sorbents concerning factors of blood coagulation and fibrinolysis; sorption capacity on the studied proteins increased in number isopropyl, butyl octyl, phenyl regardless of chemical binding ligand type; reasonably possible use include the application stage hydrophobic chromatographic fractionation of plasma technology - after salting high concentrations of ammonium salts to remove of chemicals before the final stage affinity chromatography; on the basis of the results of research asked to experimental study of the properties of the ligands silica sorbents triazyne active dyes under conditions of high hydrophobicity solution to identify effective methods of protein fractionation factors; registered technology "method of allocation of factor VIII coagulation" (0616U000066 from 27.01.2016 g.); Ukraine received a patent for utility model number 107509 "Method for allocation factor VIII clotting" (Bul. 11, 10/06/2016 p.). Degree of implementation: acts implementations, reports at international scientific forums, publications in scientific literature. Registered technology card. Areas of application: blood centers, chemical and pharmaceutical companies, research organizations. Efficiency: implementation of results of the planned research will improve previously established technology of diagnostic and therapeutic protein drugs with utilized fractions processing of donor blood plasma using previous PEG- and salt fractionation, biospecifical chromatography on macroporous silica sorbents, solvent-detergent and thiocyanate inactivation methods of viruses input stage (stages) hydrophobic chromatography. It will increase process efficiency, reduce time spent on the implementation of technology to minimize loss of activity of investigated enzymes in the process of isolation and purification.
