Komisarenko Institute of Endocrynology and Metabolism of the National Academy of Medical Science of Ukraine" popup.description2 The object of research: biochemical mechanisms regulating the function of the thyroid gland and the adrenal glands, proliferation and apoptosis in the endocrine tissues. Objective: To study the role of messenger systems (at the receptor, cellular and intra-nuclear levels), and the participation of nuclear transcription factors in mediating the regulatory influences in the tumor cells of the thyroid gland the and adrenal glands. Methods and equipment: electrophoresis of proteins; Western blot analysis, cell survival; centrifuge "Micro 22R", dropper dispensers, electronic scales "SPU 123" portable electronic scales "KERN 440-47". Results: The study of the molecular mechanisms regulating adrenocortical function at the level of nuclear transcription factors. Messenger mechanisms providing studied the for the transfer of regulatory signals of ethanolamine, N-acylated with mixture of unsaturated fatty acids in the adrenal cortex were studied. The role of transcription factors c-fos and c-jun in the transfer of N-acylethanolamines signals is very important: a level of c-fos as well as c-jun causing the initiation of cell proliferative processes, increases under compound effect. Under administration of synthetic analogue arahidonoil etanolamin – metanandamide into incubation medium its effect on the level of transcription factor c-jun in rat adrenocorticocytes was evaluated. Obtained our data about the NAE influence on proapoptotic Bax protein expression and intensity of DNA fragmentation allow its attribution to factors regulating the apoptotic processes in adrenocorticocytes. The effect of anticancer drug paclitaxel (Ptx) on biochemical mechanisms that regulating apoptosis in cells of anaplastic carcinoma of the thyroid gland was studied by us. It was shown that side by side with the apoptotic process, Ptx induces the signaling cascades that involve the NFkB nuclear factor. The obtained data of IKK1 activation and a corresponding decrease of the NFkB–IkB alpha natural inhibitor, which in one’s indicates the NFkB activation by uncanonical way. The cytotoxic effect of Ptx cells in anaplastic thyroid carcinoma is enhanced by NFkB inhibitor, dehydroxymethylepoxyquinomicin. In addition, we show the alginate capsules preserve its structure within 28 days after rat subcutaneous transplantation. Local tissue reaction of recipient appears in the form of non-specific inflammation. The results of study add the scientific data on the safety of alginate microcapsules transplantation.
