Karazin popup.description2 According to a technical task, the aim of the first stage of the project was development of novel fluorescence markers for detection and structural characterization of amyloid fibrils based on the global screening of the newly synthesized compounds. Furthermore, this work was aimed at the refinement of the qualitative diagnostics of amyloid diseases by using both the novel dyes and classical fluorescence markers, being capable of characterization amyloid fibrils of different morphology. The objects of the study are: the physical processes and mechanisms of the formation of the fluorescence response of the novel probes bound to amyloid fibrils and under the influence of various environmental factors. Absorption, fluorescence spectroscopy and computer modelling were the methods of the study. A wide range of the novel dyes, belonging to various classes, including benzanthrones, squaraines and cyanines, have been tested for their potential to be employed as markers for medical diagnositcs of amyloid diseases. 14 probes showed better sensitivity to fibrillar lysozyme and insulin than Thioflavin T. Investigation of the effect of physico-chemical parameters of the environment on the spectral properties of the potential amyloid tracers allowed us to determine optimal conditions for the employment of the novel dyes. Based on the F?rster resonance energy studies in the system of classical amyloid dyes Thioflavin T and Congo Red, the novel method for amyloid detection has been proposed. The advantage of the designed method is its ability to recognize the false-positive response of Thioflavin T, possessing high sensitivity to lipids and bacteria. Finally, the disruption of the H-aggregate absorption bands of the novel cyanines was observed in presence of the lysozyme fibrillar species, thus reporting the presence of the fibrils in the sample. This technique could be employed for the in vivo studies, because of the long-wavelength absorption and fluorescence maxima of the dyes, preventing overlapping of the dye spectra with those of the biological samples.
