There is evidence that the cause of atherosclerosis is the appearance in blood of autoantibodies to low-density lipoproteins (LDL). Immune complexes accumulate in subendotelian of some arteries, including the coronary, lead to the development of the inflammatory process and induce atherogenesis. To play an antigen-induced model of atherosclerosis, we used 80 nonlinear white rats aged 8-10 weeks. Native human LDL was injected intradermally in incomplete Freund's adjuvant of a single dose of 200 mcg regardless of body weight (Menshikov I., Fominа K., 2012). Histologic treatment of fabrics was carried out according to standard methods and stained hematoxylin-eosin, orsein. Dendritic cells identified using the marker S-100. In the study of the intima of the coronary arteries by 8-10 weeks after immunization, there was a gradual thickening subendothelial layer, and increasing the degree of lymphocytic infiltration. Dendritic cells in small amounts were detected in the subendothelial layer. 12th week was characterized by a more marked lymphocytic-gistiocytic infiltration and migration of smooth muscle cells from media to intima. Revealed moderate lipoids with zonal edema. Dendritic cells formed clusters and increased the degree of their interaction with other cellular elements. After the 16th week was some local damage to the endothelium of the coronary arteries, greatly increased the number of cells with numerous lipid droplets in the cytoplasm (so-called. foam cells). Dendritic cells were detected in all layers of the vessel wall. Distinct changes in the structure of the intima of the coronary arteries we observed at 18-20 weeks in the form of a large number of foam cells, as well as phenomena of liposclerotic. Dendritic cells in this period experienced ultrastructural changes. Thus, by using the antigen-induced model of atherosclerosis in rats managed to obtain the characteristic of atherogenesis-cellular restructuring of the walls of the coronary artery and to define the role of dendritic cells in the atherosclerotic vascular damage. The object of study refers to the histology, namely, the structure of the wall of the coronary artery in experimental atherosclerosis. The purpose of study was to establish characteristics of the interaction of dendritic cells with the microenvironment in the composition of the internal organs in preclinical and clinical stages of the modelling pathological conditions. Research methods: histological, immunohistochemical, electron microscopy, morphometric, biometric methods, statistical analysis. This paper studied the structural, ultrastructural, immunohistochemical and quantitative characteristics of dendritic cells and their microenvironment, how they interact at various stages of experimental atherosclerosis. Also identified stages of preclinical and clinical changes of the immune, stromal and epithelial components of the walls of the coronary vessels. The results of the thesis will provide the opportunity to form a theoretical basis for the use of immunomodulation as a component of interventions for the prevention of atherosclerosis and also as a component of its treatment. The results can enter into the cycle of lectures for students and doctors on histology, pathological anatomy, pathological physiology, special topics of clinical medicine.
