The goal - comparative molecular genotyping of F.tularensis strains, isolated from different sources in the natural foci of tularemia in Ukraine, to study in comparative perspective in vitro and in vivo their phenotypic individual properties, to determine the epidemiological features of disease tularemia in people. Methods: bacteriological, biological, cultural, cytomorphological, molecular, immunological, environmental and epidemiological, clinical, statistical. On the basis of multylocus-VNTR analysis molecular genetic typing of 199 strains of F. tularensis holarctica was conducted. The strains were isolated in different landscape-geographical zones (Woodlands, wood-steppe, steppe). Their genotypic diversity was detected, 44 genotypes were identified of different A, B, C groups. Some territorial restriction was shown. Genotyping of F.tularensis strains was performed by determining of the multiplicity of 4-VNTR loci repetitions with comparative frequency characteristic of alleles of polymorphic loci VNTR-FT-M3, FT-M6, FT-M19, FT-M20 finding. Genotyping of 9 strains as natural isolates isolated in the Odessa region from ticks in the 2015-2016, them identical allelic profile was found in 4-VNTR loci studied, resulting in all of them attributed to genotype V111, exept two strains: № 108 (2015 g.) and №60 (2016). In comparative perspective on in vitro models of human peripheral blood cells (PBL) and in vivo in white mice biomodels the properties of virulent strains of F. tularensis 38 different groups of genotypes (A, B, C) and vaccine strain F. tularensis 15 Ai were studied. There were established under the influence of natural isolates in vitro most pronounced destructive changes in macrophages, which are the target cells for genotype strains A, and neutrophyles - target cells for vaccine strain and genotype strain groups B and C. Our studies allowed for the first time to establish that the development of experimental tularemia infection in vitro makes tularemia pathogen able to adsorb platelets, activate them and encourage interaction with different F. tularensis cells PBL, which increased their cytodestruction. A correlation of the results of the comparative study of F.tularensis strains virulence using two experimental models in vitro and in vivo determined that the earliest cytodestruction, severe damage to PBL cells and parenchyma white mice organs resulted in strains of F.tularensis group A genotypes isolated from various sources, including those from people during epidemical complications, indicating their high virulence. The results of experimental studies conducted in vitro PBL cell system to determine F.tularensis strains virulence may be one of the criteria for assessing the activity of natural foci of tularemia and forecasting possible epidemiological complications. The research allowed studying basic biological properties of regional F. tularensis strain, determining the features of their genotypic and phenotypic characteristics and importance in the epidemic process of tularemia infection. Retrospective analysis of the incidence of tularemia in Ukraine found insufficient timely and accurate diagnosis of tularemia infection. Vaccinations, revaccination against tularemia in "epidrisks' groups" in enzootic areas were not performed, which was threatening factor for this infection. The analysis of incidence can serve as a basis for further planning of detailed molecular-genetic and ecology-epidemiological survey of tularemia natural foci to conduct timely preventive and anti-epidemic measures.
