Purpose: To synthesize new sorbents based on macroporous silica-inorganic matrix and hydrophobic ligands suitable for use in the process of obtaining protein preparations of plasma using the previous methods of fractionation, affinity chromatography and antiviral treatment. Methods: The methods of chemical synthesis, chromatographic methods of isolation and purification of proteases, methods of fractionation of plasma, centrifugation, affinity and ion-exchange chromatography, gel filtration, dialysis, spectrophotometry, methods for determining the activity of proteases, ELISA, electrophoretic methods, viral inactivation. As a result of the work received the following scientific and practical results: In the experiment, a series of derivatives of amino and hydroxypropyl-diasorbates with hydrophobic ligands (isopropyl butyl, octyl, phenyl) and proved their suitability for purification of factor VIII, factors of the prothrombin complex (II, VII, IX, X) and fibrinolysis (plasminogen, plasmin), removal of virus-activating agents; optimal conditions for the use of synthesized hydrophobic sorbents (pH, ionic strength of sorption and desorption, presence of polar or nonpolar solvents, chelators, etc.) are determined for efficient separation of investigated proteins; the efficiency of the combination of hydrophobic and affinity chromatography on silica sorbents has been experimentally proved for the removal of antiviral agents from solutions of protein preparations in the process of their technological preparation; grounded places of application of stages of hydrophobic chromatography in the technology of fractionation of blood plasma: after healing high concentrations of ammonium sulfate; for the removal of viral-activating chemical reagents; before the final stages of affinity chromatography; it was established that under conditions of high concentration of salts sorption properties of siliceous sorbents with ligand-active triasinic dyes change due to exposure of new hydrophobic sites to the surface of target proteins; experimental studies prove the expediency of using hydrophobic chromatography to separate inactive and active forms of blood coagulation and fibrinolysis; for the separation of prothrombin from active thrombin, the advantage over the main indices among the hydrophobic sorbents studied is better to show phenyl derivatives of macroporous silica. Degree of implementation: acts implementations, reports at international scientific forums, publications in scientific literature. Areas of application: blood centers, chemical and pharmaceutical companies, research organizations. Efficiency: implementation of results of the planned research will improve previously established technology of diagnostic and therapeutic protein drugs with utilized fractions processing of donor blood plasma using previous PEG- and salt fractionation, biospecifical chromatography on macroporous silica sorbents, solvent-detergent and thiocyanate inactivation methods of viruses input stage (stages) hydrophobic chromatography. It will increase process efficiency, reduce time spent on the implementation of technology to minimize loss of activity of investigated enzymes in the process of isolation and purification.
