Development of biosensor methods of arginine analysis in wine by using arginine-hydrolyzing recombinant enzymes. popup.stage_title Head Sibirny Andriy, Registration Date 01-02-2018 Organization Institute of cell biology, National Academy of Sciences of Ukraine popup.description2 As a result of the work in 2017: the preparative amounts of microbial cells - over-producers of human liver arginase (natural and (His) 6-tagged forms), bacterial arginine deiminase (ADI), as well as yeast alcohol oxidase and flavocytochrome b2, were obtained; the preparations of the corresponding enzymes were isolated and characterized, preparations of ADI and arginase were transferred to IMBiG NAS of Ukraine; original amperometric bioelectrodes were created and investigated, these electrodes were based on oxidases, immobilized on nano-particles (NPs) of noble metals, which were obtained by different methods (chemical or "green" synthesis); the proposed bioelectrodes for glucose assay were demonstrated to have optimized analytical characteristics compared with bioelectrodes formed with the usage of enzymes without NPs, namely, enhanced stability, extended linear range and higher sensitivity to the target analyte; the developed biosensors for glucose determination have been successfully tested on real samples of wines compared with reference methods and manufacturer data. On the base of scientific results, 6 articles, 3 chapters in books and 5 theses were published. The work was carried out in full according to the plan.
