To set up the Agrobacterium-mediated wheat transformation in vitro a modified medium (MS with addition of 0.5 mg/L BAP and 0.16 mg/l of picloram) for regeneration (up to 60%) was developed; As explants, the callus derived from immature embryos and apical meristems of three-day aseptic seedlings (18-days) was used; the concentration of antibiotics timentin (350 mg/l) and ceftriaxone (400 mg/l) was established, necessary for the elimination of agrobacteria, enhancement of regeneration, stimulation of growth of wheat plant regenerants, growth of their biomass and rooting; the histological features of wheat regeneration in the presence of the antibiotic cetifriaxone in the culture medium (regeneration was through direct organogenesis) was studied; Depending on the design, paromomycin (100 mg / l) or phosphinothricin (5 mg/l) were used as selective agents. For the first time transgenic wheat plants of cultivars Podolanka and Zimoyarka, resistant to phosphinitricin herbicide (Basta®) and expressing the gene bar, were obtained. The transformation frequency was 1.8% for the Podolanka variety and 1.6% for the Zimoyarka variety. As a result of Agrobacterium-mediated transformation of wheat in planta kanamycin-resistant and phosphinothricin-resistant wheat plants were received. The Agrobacterium-mediated and biolistic transformed corn calli of different genotypes have been obtained and the T1 plants were resistant to the herbicide phosphinothricin (Basta®). The vectors pCB182 and pCB241, which contain the sequence of the resistance gene cry1A(b) to the larvae of insects, were created and capable to transfer the gene to the plant genome. All necessary stages of cultivation in vitro of apical meristem of spelt were worked out to obtain regenerants. Based on the successful genetic transformation of wheat and corn, the molecular genetic features of biotechnological plants have been determined and the peculiarities of the transgenes performance were studied.
