The work involved immunohistochemical and statistical methods of analysis. The aim of the study was to characterize the effect of humoral microenvironment factors on tumor expression of associated markers under various conditions of microenvironment. The objectives of the study were to cultivate MCF-7 mammary adenocarcinoma cells under various microenvironmental conditions (under the influence of IFN-?-2b, k-media from T-lymphocytes and MSCs), and also to analyze the nature of the expression of surface markers indicative of phenotypic changes in tumor cells and on the signs of their differentiation. As a result, it was possible to obtain histological preparations of multicellular tumor spheroids from the MCF-7, HepG2, HT29 cell lines. Microhistological analysis demonstrated the characteristic structure of microspheroids: a thin surface layer of actively proliferating cells, an inner layer of cells at rest, and an inner zone of necrosis. With the help of immunocytochemistry, it was found that the most active secretion of proteins by tumor cells occurs in the surface layer. The highest expression of vimentin, E-cadherin and Pan-cytokeratins occurs precisely in the surface layer of the microspheroid. It was demonstrated that K-media from MSC, as well as IFN-?-2b stimulate the synthesis of tumor cells by cytokeratins 10, 13, 14, 15, 16, and 19, which promotes the appearance of cells with a more differentiated phenotype. At the same time, incubation of tumor cells with K-medium from T-lymphocytes somewhat reduces the secretion of cytokeratins 5, 6, 8 and 18, and with IFN-?-2b - almost blocks. But incubation with K-medium from MSC stimulates the expression of these proteins. The amount of rEFF+ cells increases under the influence of the test substances, but most significantly when incubated from the c-medium from T-lymphocytes and MSC, by 22.2% and 16.7%, respectively. The decrease in E-cadherin expression under the influence of K-medium from MSC and IFN-?-2b indicates the plasticity of tumor cells that can change phenotypic signs under the influence of microenvironment conditions. It was also shown that the K-medium from T-lymphocytes stimulates the expression of estrogen receptors, which is a promising way in the selection of antitumor therapy, as it increases the population of cells sensitive to the action of drugs. Scientific and practical significance. Thus, as a result of the work, it was confirmed that the investigated substances (IFN-?-2b, k-medium from T-lymphocytes and MSC) to varying degrees contribute to a change in the phenotype of tumor cells, enhancing the properties of differentiation, and also enhance the expression of estrogen receptors, which is promising for anti-estrogen therapy.
