The aim of work - to study the expression of с-MYC gene in CLL patients depends on radiation anamnesis, mutational status of NOTCH1 and IgHV genes. Methods: clinical, polymerase chain reaction, direct sequencing of reaction products, statistical. Equipment: thermocycler Bio-Rad C1000 (N10450137), dispensers. The main results: it was proposed the method for detection of mutations in non-coding 3'UTR region of NOTCH1 gene using polymerase chain reaction with followed restriction. It was established that the frequency of such mutation was low (2.5 %) and did not differ in CLL patients depend on radiation anamnesis. Association of the frequency of NOTCH1 gene’s mutations in non-coding region and in the exon 34 with structure of B-cell receptor was found. It was maximal when unmutated IgHV genes of I phylogenetic clan expressed; lower - at expression of IgHV1-69 gene and minimal at expression of the other unmutated IgHV genes. Expression of с-MYC gene in lymphoid cells of peripheral blood in CLL patients was low and does not correlate with mutational status of IgHV genes, mutations of NOTCH1 gene, rs6983267 and rs2456449 polymorphisms in non-coding region of 8q24 chromosome near с-MYC gene, radiation anamnesis of patients, but in CLL patients with secondary solid tumors (р = 0.02). Field of application: radiobiology, hematology. Publications: 3 papers, the patent.
