Objects of the research: standard mammalian cancer cell lines; original human cell lines, obtained in the department of human genetics, ICR mice. The aim of the study – research of the biological active substances (cytokines, lectines, steroid hormones, inhibitors) on the gene expression of reparative enzyme MGMT and presence of it protein in cancer and normal human cells in vitro. Cellular, cytogenetic, molecular-genetic, immunochemical, bioinformatic and statistical methods were used. For the first time the statistical reliable correlation between of the expression of the MGMT gene and the level of mutational variability at the chromosomal level was revealed during the establishment of the cell line 4BL and stress-factors impact. By in silico method presence of the MGE and numerous regulatory sites in the promoter of MGMT gene was shown. The spectrum of growth factors, cytokines and lectins as potential regulators of MGMT gene expression has been expanded; dependence of their influence from the protein concentration, investigated drugs composition, cell lines features, was revealed. For the first time we shown that estrogen in physiological concentrations is able to activate the MGMT gene expression at the mRNA and protein level in different human cell lines. New rational method for the detection of the new inhibitors of MGMT enzyme activity has been developed. The study is fundamental since its results deepen understanding of the influence ways on the expression of MGMT gene and the activity of it protein in order to correct genetic damages. Subsequently, obtained results may be a basis for development of approaches to optimizing chemotherapy of cancer with the use of alkylating compounds.
