Information × Registration Number 0219U100239, 0116U000333 , R & D reports Title Search of compounds with anti-diabetic properties which direct to restore the tissue regulation of energy balance at the presence of type 2 diabetes mellitus with overweight or obesity popup.stage_title Head Poltorak Viktoriya V., Доктор медичних наук Registration Date 31-01-2019 Organization State Institution “V.Ya.Danilevsky Institute of Endocrine Pathology Problems of Academy of Medical Sciences of Ukraine” popup.description2 The object:were rats,mice,condensed and spiro- nitrogen- and sulfur-containing heterocyclic compounds.The purpose:was to identify among the group of new derivatives of nitrogen and sulfur-containing heterocycles the compounds that able to inhibit the development of determinant pathogenic units of type 2 diabetes mellitus (DM)(insulin resistance and associated metabolic displacements, obesity, fatty liver disease,pancreatic b-cell dysfunction), due to the influence on tissue regulation of energy balance.Methods:organic synthesis,physical and chemical analysis,biochemical,physiological,pharmacological, immunological,histological.Methods of synthesizing new pyrazolo[3,4-b]pyridine-5-carbonitriles,pyrazolo [3,4-b]quinolin-5-ones,dihydrospiro[indoline-3,4'-pyrazolo[3,4-b]pyridine]-2,6 '(5'H)-diones, keto acids from the group of 3-spiro-pyrrolizinyl-2-oxindoles,3-(2-amino-4-arylimidazol-5-yl)-propionic acids,pyrrolo[1,2-c]imidazoles,dispiro[indole-3,2'-pyrrolidine-3',5''-[1,3]thiazolidine]-2,4''(1H)-dione,dispiro[acenaphthylene-1,2'-pyrrolidine-3',5''-[1,3]thiazolidine]-2,4''-diones and N-methyl-b-(2,4,6-trioxohexahydropyrimidin-5-yl)arylalanines with potential antidiabetic activity have been worked out.The analytical attestation of substances AD-1408,SІ-630 and micronized form of the compound LV-2418 has been carried out.In rat models of type 2 DM with obesity the complex antidiabetic properties of four substances,which affect energy metabolism (promising sirtuin-1 activator SІ-630,micronized form of promising 11b-HSD1 inhibitor LV-2418,fatty acid receptor agonists K-1 and K-2),have been evaluated.The prospects of further studies of the complex antidiabetic properties of the original compound SI-630,which not only influences key pathogenic units of DM,but also reduces the risk of premature loss of mitochondrial function,pancreatic cells and the development of non-alcoholic fatty liver disease,are justified.The field of application:healthcare,pharmindustry.
