Karazin Kharkiv National University popup.description2  According to the technical task of the project, quantitative parameters of the amyloid fibril growth (lagtime, rate constant, Thioflavin T fluorescence response), as well as binding parameters of the potential inhibitors of fibrilogenesis with different protein types (native protein, oligomers, amyloid fibrils) have been determined. Furthermore, effects of modulators on the amyloid nucleation and elongation have been differentiated, and protein binding sites for the inhibitors have been suggested. Next, the most effective and specific inhibitors of the amyloid formation process of different proteins have been selected, including those capable of retarding the pathogenic protein aggregation on the membrane surface. Finally, molecular mechanisms of the modulation of the membrane structural and functional state by the potential anti-amyloid drugs, as well as mechanisms underlying their effects in aqeous solution, have been proposed using the results of the molecular dynamics simulations. All results obtained are of great importance for the directed applications in biomedical research.
