Aim: to study the mechanisms of modification of structural parameters and the metabolic state of cord blood nucleated cells and donor red blood cells under the influence of various cryoprotective solutions and low temperatures. In the work modern research methods were used: flow cytofluorimetry, confocal microscopy, methods of electrophoresis in PAAG, small-angle light scattering and spectrophotometry. In results of the study it was found that the use of cryoprotective mixtures containing DMSO at a concentration of 7.5 or 10% and ascorbic acid at a concentration of 0.1 and 0.15 mM or N-acetyl-L-cysteine at a concentration of 10 and 15 mM for cryoconservation of cord blood nucleated cells, including hematopoietic progenitor ones, reduced the amount of cells contenting ROS excess. This allows saving a larger number of safe and viable cells at defrosting and modeling transfusion stages. The most pronounced cytoprotective effect was observed in samples cryopreserved with N-acetyl-L-cysteine, which allows preserving up to 80% of cord blood nucleated cells and up to 85% of hematopoietic ones in a viable state. On the basis of the conducted research a new method of cryopreservation of cord blood nucleated cells for clinical practice has been developed. It has been also revealed that structural modifications of membrane proteins during freezing largely correlate with the effectiveness of their cryoprotectant actions and affect the mechanical stability of human red blood cells. The ROS formation in red blood cells under the influence of cryoprotectants indicated their possibility to affect the cell stability during cryopreservation.
