Based on preliminary bioinformatics search and analysis of α-, β-tubulin and actin genes encoded in the genomes of A. thaliana, rice (O. sativa), long-flax (L. and sitatissimum), potato (S. tuberosum) and tomato (S. lycopersicum), primers were selected for the analysis of intron length polymorphism within each species. This year, the synthesis of species-specific PCR primers was carried out to assess the polymorphism of the length of introns of the genes of the key proteins of plant cytoskeleton, namely α-, β-tubulin and actin. The conditions for the amplification of the 1st intron of α-tubulin A. thaliana, rice (O. sativa), flax (L. and sitatissimum), potato (S. tuberosum) and tomato (S. lycopersicum) using the appropriate species-specific primers were selected . In the case of β-tubulin genes, for each model plant species, two pairs of primers were synthesized, allowing to estimate the length polymorphism of both the first and second introns. Optimal conditions were selected for amplification of introns of β-tubulin genes in A. thaliana, O. sativa, L. and sitatissimum, S. tuberosum and S. lycopersicum. The synthesis of species-specific primers was carried out to assess the polymorphism of the length of introns of actin genes in model species of higher plants. Optimal conditions were selected for the amplification of actin intron genes in A. thaliana, O. sativa, L. and sitatissimum, S. tuberosum and S. lycopersicum species. A preliminary PCR analysis of interspecific and varietal samples of model plant species was carried out using new developed species-specific DNA markers.  Product Description popup.authors Blume Yaroslav B.
