The preparation of the conjugates was tested on immunoglobulin G (IgG) and included modification of IgG 2-iminothiolane and modification of PerCP or tandem dyes by succinimidyl-4- (N-maleimidomethyl) cyclohexane-1-carboxylate (SMCC) conoxylate (SMCC) fluorescence labeled antibody (IgG) antibody - PC5, R-PE-ISC-650 or PerCP. It was found that increasing the number of cross-linkers used in protein modification leads to the formation of unwanted fluorescent labeled IgG aggregates and insoluble precipitate. The decrease in the number of cross-linkers used results in only partial labeling of antibodies or the formation of IgG conjugates with a small number of fluorescent label molecules. The fluorescence of all the conjugates obtained is well excited by a 488 nm laser, but in the case of labeling with antibodies by tandem dyes (PC5, R-PE-ISC-650) for use in cytofluorimetric studies, it is necessary to obtain the conjugate at a ratio of 1: 1 to tandem dye, whereas to obtain a similar signal for conjugates with PerCP it is necessary to obtain a conjugate in the ratio of 1: 4-5 the number of antibodies to PerCP.
