The object of a study is the process of immobilization of GST fusion proteins on the surface of modified GNPs and influence of the conjugated proteins on the sensitivity of tumor cells to chemotherapy. The goal of the present work is to create a molecular system for the delivery of specific proteins to tumor cells, based on a study of interaction of GNPs with proteins of different classes for the further development of antitumor therapy. Research methods - chemical reduction, surface reactions, optical spectroscopy, standard molecular procedures, purification of GST-fusion proteins, western blot analysis, human cell cultures, transfection of human cell lines, endocytosis, fluorescence microscopy, statistical methods. A novel method of protein delivery into cells was developed, i. e, the GST-fusion proteins of a MRPS18 family (S18-1-3), TP53 and RB were immobilized on a surface of GNPs (of the 5 nm diameter) and delivered to human tumor cells (MCF7, PC3, and PNT2) by pinocytosis. Upon simultaneous introduction of the S18-2 and TP53 proteins the synergy of apoptosis caused by the protein TP53 was observed. The presence of GST fusion proteins significantly reduced the resistance of PC3 and PNT2 prostate tumor cells to anticancer drugs, such as vincristine and doxorubicin. Thus, GST fusion protein conjugates immobilized on the surface of GNPs increase the sensitivity of malignant cells to anticancer therapy, which can be used to develop anticancer drugs.
