Subject of the study: morpho-functional changes of epidermis, dermo-epidermic junctions, dermis and appendages of the skin. Purpose of the study: experimental study of morpho-functional damage of skin barrier properties in rats under a long-term intervention of compounds of heavy metal NPs (Pb, Cu, Ag та Fe) and determination of mechanism of their resorption through the skin. Study design: toxicological, histological, histochemical, submicroscopic, luminescent and polarization microscopy, as well as morphometry and variation statistics methods. Results and their novelty: It is stated for the first time, that long-term effect of metal NP compounds on the uninjured skin leads to morphological changes, which are seen at microscopic and submicroscopic levels in all skin layers of epidermis, dermis and appendages. All changes have signs of destruction and point to the skin barrier function damages. Submicroscopic changes of membrane and cytoplasm of corneocytes of the stratum corneum and keratinocytes of all layers, as well as changes of the basal membrane structures and hair follicles and holocrine glands determine main ways of metal NP resorption through the uninjured skin of rats under their long-term intervention. Mechanisms of intracellular, transcellular and intrafollicular transport play a significant part. The results prove that not only toxic properties are of great importance in providing mechanisms of resorption through the skin of metal NPs, different by their physico-chemical parameters, but, also, their size, type, concentration and time of their application onto the skin.
