First of all, we have experimentally developed and validated the optimal DNA extraction protocol, which is critical for the success of subsequent analysis and adequate data interpretation. Based on DNA quality and quantity data we recommend to use DNeasy Power Soil KIt (QIAGEN) commercial kit. This kit gives quick and high-quality results and can be used without any specific equipment and with limited lab space. Thus, such protocol is the optimal choice for field work at Vernadsky Research Base. Another important issue addressed by our project was the development of optimal primer set and amplification protocol for specific taxonomic analysis. Monitoring studies are based on well-developed and thoroughly validated methods, which yield relevant and comparable results in long-term perspective. Therefore, we tested the optimal PCR protocols and primer annealing temperatures for our type of samples. Our results were different from those provided by the previous studies, which highlighted the importance of protocol validation in each particular case. Taking into account the validity of our taxonomic analysis results and the high performance of methods, we recommend to implement the protocols provided by us in the course of marine monitoring program at Vernadsky Research Base.
