Both experimental compounds were shown to induce apoptosis in glioma cells, but no evidence of oxidative stress was observed. Adding of a non-toxic dose of menadione to the above-mentioned cell lines in vitro resulted in a pronounced synergistic effect, significantly (1.5 times) enhancing the cytotoxic activity of 4-thiazolidones. This phenomenon was also accompanied by significant depolarization of mitochondria and a sharp increase in superoxide anion production in mammalian glioma cells. In the second stage of this project, synergistic effect of small doses of several celandine alkaloids and traditional anticancer drug doxorubicin was investigated towards mammalian cells. It was shown that the therapeutic effect of doxorubicin was enhanced only by the sanguinarine, which leads to oxidative stress in the target cells, whereas structurally related chelidonine (acting on the cytoskeleton) does not have this effect. The synergistic action of sanguinarine and the anticancer drug doxorubicin was most pronounced in human drugh-resistant leukemia cells, which are characterized by overexpression of ABC transporter proteins. Thus, the use of low-dose prooxidants allows to enhance the cytotoxic action of anticancer drugs, which are unable to induce ROS, against human malignant cells and may have practical implications in clinical practice.
