he object of study - the spatial structure of the factors of translation elongation.
The aim of the work is to reveal for the first time the spatial organization of human elongation factor eEF1Bα in and to determine the structural regions responsible for the multimerism of the eEF1Bβ protein.
Research methods - molecular biological (PCR, cloning, mutagenesis, expression of recombinant proteins), physicochemical (analytical ultracentrifugation, HDX-MS), biochemical (gel electrophoresis , chromatographic isolation of individual proteins, Western blot), bioinformatics (molecular modeling).
A comparative analysis of previously obtained and new data of the structural dynamics and multimeric state of eEF1Bα and X-ray diffraction analysis of the domains of this protein.
A spatial model of the eEF1Bα structure was constructed.
The level of eEF1Bβ multimerization by the method of analytical ultracentrifugation (AS) in the speed and equilibrium modes was studied.
The regions of eEF1Bβ responsible for multimerization were identified by analysis of deletion mutants.
