The sandwich-IEA test-system based on immobilization of antibodies on ammonium perchlorate-functionalized polystyrene surface for quantification of free PSA has been developed. A comparative analysis of the effects of polystyrene chemical modification with ammonium perchlorate on a sorption of the recombinant HSP-60 proteins with different tags was carried out. It was shown that the most effective method used the functionalization of polystyrene surface with ammonium perchlorate (relative degree of hydrophilization 4.8) and recombinant proteins rHSP-60-GST or rHSP-60-His6 (at concentration of 5 μg/ml or more), that allowed to increase the IEA signal by about 1.5 times and positivity index by 30% without reducing of nonspecific interaction level. The developed technique of polystyrene surface functionalization can be used in diagnostic systems for biomedical purposes. The developed test-system for quantification of free PSA can be used to diagnose and monitor the effectiveness of prostate cancer therapy. The IEA technique for detecting IgG-specific antibodies to HSP-60 can be used in development of the test-system for urogenital chlamydia diagnosis.
