The aim of this work is to study the effect of a mutation in the BRCA1 gene on the development of genomic instability in human peripheral blood lymphocytes. Research methods: molecular genetic - the method of polymerase chain reaction and electrophoresis for the separation of the amplification PCR - search for mutations 5382insC, 185delAG, 300T> G, 4153delA in the BRCA1 gene; Comet assay method. Individuals carriers of the 5382insC mutation in the BRCA1 gene have been identified. A comparison of the level of genomic instability in peripheral blood lymphocytes of conventionally healthy individuals and persons carrying a mutation in the BRCA1 gene has been carried out.The features of the development of genomic instability in persons carrying the 5382insC mutation in the BRCA1 gene have been revealed - the presence of a cell pool with a high level of DNA damage on the background of the absence of an increase in the level of chromosomal aberrations and activation of the checkpoint - control at the S stage of the mitotic cycle. Apoptosis is the key mechanism for regulating the development of genomic instability in lymphocyte cultures of individuals who are asymptomatic carriers of the 5382insC mutation in the BRCA1 gene.
