Computational algorithms and methods for molecular docking and molecular dynamics simulations of human cytosolic leucyl-tRNA synthetase (HscLeuRS) and Rag GTPases, for analysis of stable states of HscLeuRS, RagA/RagC, RagB/RagD were selected. The HscLeuRS purification scheme has been modified. RagA and RagC GTPases were expressed in E.coli BL21(DE3)pLysS cells. Expression of LARS1 and SESN2 genes, which encode HscLeuRS and Sestrin2, respectively, was studied by real-time quantitative PCR in HEK293T cells under conditions of norvaline treatment.
