The presence and degree of specific binding of AIMP1 / p43 and EMAP II proteins to cyclodextrins and dextran-70 were investigated by fluorescence spectroscopy and light scattering. The stability of preparations of recombinant proteins AIMP1 / p43 and EMAR II, as well as their complexes with increasing temperature was tested. The experiments revealed a significant stabilization of the AIMP1 / p43 and EMAP II protein molecules in solution, which makes it possible to obtain highly concentrated preparations for further structural and functional studies. The analysis of exposure of aromatic amino acid residues in the structure of recombinant cytokine EMAP II was performed. t was found that γ-CD specifically binds to EMAP II, with the most probable binding site being the partially exposed Trp125 residue located in the recess on the surface of EMAP II next to the functionally important lysine-rich cluster. The aggregation properties of the cytokine EMAP II have been studied and it has been shown that the polysaccharide dextran 70 inhibits protein aggregation processes with increasing temperature. A possible mechanism of aggregation of the recombinant cytokine EMAR II is proposed. Temperature stabilization of the antitumor cytokine EMAP II is shown.
