Research object – Spatial organization of the translation elongation complex eEF1B and its subunits.
Purpose of work – To reveal for the first time the spatial organization of translation elongation complex eEF1B and its subunits/
Research methods – molecular biological (PCR, cloning, mutagenesis, expression of recombinant proteins), physico-chemical (HDX-MS, analytical centrifugation,X-ray scattering), biochemical (chromatographic purification of individual proteins, Western blot), bioinformatics (molecular modeling).
Results and novelty:
For the first time the structural dynamics of eEF1B, eF1Bβ and eEF1Bγ subunits of multiprotein complex eEF1B was elucidated experimentally. For the first time molecular form of eEF1B, eEF1Bγ and trimeric eEF1Bβ proteins was determined. For the first time experimenatally based computational models of spatial organization of eEF1B, eEF1Bβ and eEF1Bγ were generated. For the first time atomistic model of spatial organization of multiprotein complex eEF1B was created.
