During the project implementation the methodology of complex processing of hydrobionts of the Antarctic region on the example of Antarctic jellyfish was developed, which allowed to make maximum use of raw materials and consistently obtain serine protease fraction, collagen, further hydrolyzed to low molecular weight fragments and peptide fraction with peptide fraction. The serine protease fraction was obtained from the lyophilisate of Antarctic jellyfish biomass dissolved in the appropriate buffer. The precipitate after the biomass lyophilisate centrifugation step and the fraction that did not bind to the affinity sorbent in the serine protease step were used to produce collagen. The material for obtaining the peptide fraction was waste obtained at all stages of collagen extraction and serine protease fraction. Enzyme-electrophoretic analysis of the serine protease fraction revealed the ability of enzymes from Antarctic jellyfish to efficiently break down collagen, gelatin and to a lesser extent fibrinogen. Relatively high proteolytic activity of serine proteases was found, evaluated using casein as a substrate. The use of synthetic low molecular weight substrates showed higher activity relative to the substrate H-D-Phe-Pip-Arg-pNA and complete lack of activity when using N-α-benzoyl-DL-Arg-pNA. It was found that the fraction of serine proteases from Antarctic jellyfish contains a small amount of proteins with isoelectric points at pH 3.0 and 5.0-6.0.  Product Description popup.authors Vovk Tetyana B.
