Positive controls in the form of plasmids with both normal and mutated sequence were synthesized. The efficiency and specificity of primers for the studied mutations were determined. The most sensitive primers to mutation A (V600E), which is the most common and clinically significant. The specificity of their work showed that they are able to detect two mutations A and C, which can be differentiated using both pairs of primers (primers A and C). Tissue and ovarian tumors of both fresh-frozen and paraffin blocks were screened. As a result, positive for A mutation samples were detected. The results were confirmed by Senger sequencing and next-generation sequencing. A protocol was developed, laboratory samples were made, and a diagnostic kit was tested to detect clinically significant mutations in the BRAF gene at the position 600.
