The present study investigated the effect of plasmin(ogen) on redox balance and signaling pathways that control the survival and migration of 4T1 mouse breast adenocarcinoma cells with different expression levels of adapter/scaffold protein Ruk/CIN85. Ruk/CIN85 was shown to play an important role in maintaining the viability and migratory activity of adenocarcinoma cells under the influence of plasmin(ogen), contributing to acquisition by cancer cells the properties of highly malignant cells. Plasmin(ogen) was found to enhance the production of reactive oxygen species (ROS) in cells with basal and down-regulated Ruk/CIN85 expression. Ruk/CIN85 overexpression maintained the redox balance, survival, and motility of 4T1 cells by activating the synthesis of the TIGAR regulatory protein required for the functioning of the antioxidant enzyme system. Ruk/CIN85 has been shown for the first time to modulate changes in the levels of apoptosis regulator proteins and the genome repair system (Bxl-xl and PARP-1) in breast adenocarcinoma cells in response to plasmin(ogen) exposure. The results expand the understanding of the molecular mechanisms underlying malignancy of breast cancer cells regulated by plasminogen/plasmin system proteins and Ruk/CIN85 adapter protein.
