Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2  The expression of the c-MYC oncogene and the level of double-stranded DNA breaks in blast cells of patients with acute myeloid leukemia (AML) were assessed considering the amount of CD34+ cells. The study was performed on bone marrow samples of 31 patients with AML. The analysis of immunophenotype showed that in 45.1% of cases of AML the presence of the CD34 marker was observed, in 48.4% of cases this marker was absent. When assessing the degree of blast differentiation based on the expression of MPO, CD117, CD11b, and CD14, it was found that in 48.4% of patients, blast cells were predominantly immature (phenotype MPO+CD117+). In 19.3% of cases, the majority of blasts were at the late stages of differentiation (phenotype CD11b+CD14+). There was a tendency towards an increase in the level of DNA damage in CD34+ -blasts compared to CD34- (16.9 ± 1.6 and 11.2 ± 2.1% tDNA, respectively). In addition, it was found that the expression of the c-MYC oncogene was associated with CD34-status and was significantly higher in CD34+-blast cells (0.92 ± 0.31 and 0.39 ± 0.09 a.u., respectively, p <0,05). An increased level of DNA damage was determined in MPO+CD117+ -blast cells compared to cells with the CD11b+CD14+ phenotype (15.5 ± 1.8 and 8.9 ± 1.6% tDNA, respectively, p <0.05). At the same time, no significant difference was found between the expression of c-MYC and the degree of blast differentiation. Further studies are needed to clarify the association of genome instability in CD34+ cells with c-MYC-mediated replicative stress and the degree of blast maturity in AML.
