Materials and methods. Cytofluorimetric, spectrophotometric, spectrofluorimetric, biochemical, immunocytochemical, histological methods, volumetry, physical and mathematical modeling, in vivo microscopy, cell culture, cryopreservation, experimental transplantation were used. Results of the research. Screening of three-dimensional structures of different chemical nature as carriers for MSC culture was performed. Chitosan matrices with a high degree of deacetylation and chitin carriers obtained after demineralization of natural sponges of the Verongida division supported normal cell adhesion and proliferation. Scaffolds based on collagen and its derivatives were safe and biocompatible for 3D culture of MSCs. To assess the reparative potential of MSCs, a modification of the full-layer excisional skin wound model in mice that prevents contraction was developed. It was shown that cryopreserved MSCs embedded in plasma-based hydrogel promoted the process of wound healing similar to non-cryopreserved MSCs. MSCs in 3D scaffolds are more sensitive to the generally accepted method of cryopreservation, which includes slow two-stage freezing under the protection of 10% cryoprotectant DMSO and fetal bovine serum, compared to cells in suspension.
