Research methods - methods of experimental oncology, methods of cell culture, the method of flow cytofluorimetry, the method of quantitative polymerase chain reaction, statistical methods. The aim is to develop technologies for obtaining antitumor vaccines aimed at eliminating tumor stem cells, and to establish the feasibility of their use in the immunotherapy of malignant neoplasms. It was found that the cells of the primary culture of non-small cell lung cancer cells in conditions of low-adhesion growth form a side population (SP), characterized by an increase in the exclusion index of the dye R-123 compared with adhesive PC (p≤0.02). According to the phenotypic characteristics of primary culture cells of non-small cell lung cancer cells in conditions of low-adhesive growth form tumor stem cells, due to increased percentage of CD90 + CD117 + cells (p = 0.01) and CD133 + cells (p = 0.02) compared to standard culture conditions. There was a significant increase in the expression of ALDH1, CD44 and CD133 mRNA (p≤0.02) in primary culture cells of non-small cell lung cancer cells in conditions of low-adhesion growth, which also indicates the presence of tumor stem cells in culture. Dendritic cells loaded with lysate of tumor cells enriched in tumor stem cells belong to mature cells, due to the increase in the number of CD83 + cells (p = 0.02). The use of lysate of primary tumor cells enriched in tumor stem cells does not have a pronounced suppressive effect on the maturation of dendritic cells, instead, a significant decrease in the expression of IDO mRNA was observed, p <0.05. LPCs derived from primary non-small cell lung cancer cell cultures enriched with tumor stem cells promote the generation of mature dendritic cells and enhance cytokine mRNA expression of cytokines with antitumor activity, namely TNF-α and TLR-4 (p).
