Description

 The combination of hyperglycemia and hyperlipidemia causes ultrastructural changes in VMH neurons in rats, which are characterized as EP stress in the form of tank expansion, increase in perinuclear space, increased apoptosis due to mitoptosis. Functionally, EP stress is characterized by a decrease in chaperone BiP / GRP78 and an increase in the content of UPR sensors ATF6, IRE1, PERK, as well as an increase in neurons BAX-dimer, caspase-3 and a decrease in Bcl-xL. Pharmacological correction with metformin, propionate and their combination improves the condition of neurons and causes some compensation of EP stress. Propionate-induced autophagy due to the formation of vesicles from EP structures demonstrates an effective antiapoptotic mechanism of counteraction to stress by the ER degradation system and can be considered as a promising area of neuroprotection in diabetes mellitus2.
