popup.description1
The aims are investigation of cumulative neurotoxic effects of multicomponent pollution by airborne PM and neuroactive pharmaceuticals, biomaterials and toxic metals using own approach on assessment of acute neurotoxicity using brain nerve terminals, in vitro and in vivo animal models; identification of types of PM that can be carriers for these pollutants; molecular mechanisms of neurotoxicity; interconnection between PM and susceptibility to SARS-CoV-2 infection; and countermeasures.
popup.description2
The diversity of interference effects of pollutants, namely heavy metals and smoke particulates, on key presynaptic processes was revealed. Reciprocal neurological hazard of multi-pollution with cadmium and lead on the one hand and smoke PM from the burning of poplar wood and medical masks on the other hand due to disturbance of excitation-inhibition balance and membrane potential of nerve terminals has been proven.
The combined application of Hg2+ and carbon-containing particles (carbon dots synthesized from citric acid/urea) as model air pollution PM showed for the first time that they were able to attenuate Hg2+-induced acute neurotoxicity in nerve terminals. It was shown for the first time that nanodiamonds did not possess such an ability, thereby demonstrating the diversity of effects of carbonaceous air pollution PM and their potential to modulate Hg2+-induced neurotoxicity.
For the first time, it was shown that smoke aerosol PM from poplar wood did not affect Hg2+ -induced changes in the accumulation and release of L-[14C]glutamate and [3H]GABA in nerve terminals that was proved by the combined effect of smoke preparation and Hg2+.
Nitrogen-doped multilayer graphene, N-MLG has no neurotoxic features and is biologically compatible in the concentration range of 0.01-1.0 mg/ml. In a biological system, N-MLG did not reduce/enhance Cd2+/Pb2+/Hg2+-induced neurotoxicity in nerve terminals.
The developed methodical approach involving the model of acute metal-induced neurotoxicity in nerve terminals can be applied to analyze the ability of various particles/compounds to have signs of heavy metal chelation in biological systems and can be used in environment monitoring.
