It was found that the most favorable for the survival and proliferation of explant buds of grape varieties Arkadiya and Zagadka was a nutrient medium with antioxidants 1.5 mg/l ascorbic acid and 0.5% glycine both under etiolation conditions and under lighting with a 12-hour photoperiod. The content of polyphenolic compounds in the green tissues of experimental explants of these variants was lower than in the control, 15 days after the introduction after the end of the etiolation period, and after 30 days the amount of polyphenols increased, which, combined with good biometric indicators of microclones, indicates optimal conditions for growth and development of microclones. Callus cultures were obtained from leaf explants of grapevines of technical and rootstock varieties on a nutrient medium MS containing 3.0 mg/l NOC and 0.5 mg/l 6-BAP. The best medium for co-cultivation of calluses was the nutrient medium MS with 2.5 mg/l IOC, 1 mg/l 6-BAP. The obtained callus cultures had an average degree of development, which indicates partial tissue compatibility. Calculations of the regeneration potential of chubuks for grafting showed that the highest this indicator was observed in two rootstock varieties Dobrynya, R*G 3562 – 18. Varieties R*R 101-14 4923, Kardishash, Odessa black – 9, the most optimal combination for grafting, taking into account the simultaneous dates of the beginning of callusogenesis and the formation of a circular callus on the sections. Rootstocks B*R СО4 1791 and P*G 5941 almost did not show regeneration properties, the sections on the surface were oxidized, which may indicate inhibition of callusogenesis due to the formation of oxidation products of polyphenolic compounds when interacting with cell enzymes and active forms of oxygen.
