The purpose of the study: Determination of biophysical characteristics and experimental substantiation of the possibility of using these characteristics as potential therapeutic targets for antimetastatic therapy Research methods and equipment: biochemical methods, cytofluorimetric method using labeled monoclonal antibodies, IR and Raman spectroscopy, confocal and CARS microscopy. Theoretical and practical results: Spectral markers of highly metastatic (LLC) and low metastatic (LLC/R9) lung carcinoma cells under the condition of their adhesive and deadhesive growth were determined. The total lipid fraction isolated from LLC and LLC/R9 tumor cells was analyzed using IR spectroscopy. LLC and LLC/R9 cells were visualized under the condition of their adhesive and deadhesive growth using confocal microscopy and CARS microscopy. Changes in the number of live LLC and LLC/R9 cells were studied during incubation of cells for 3 days of their adhesive and deadhesive growth (experimental model of circulating metastatic cells). A comparative analysis of LLC and LLC/R9 growth kinetics under different growth conditions was performed using mathematical modeling and the optimal incubation period of cells for further studies was determined. The number of live cells, the level of apoptosis, the distribution of cells by cell cycle phases (survival characteristics), and the mesenchymal characteristics of membrane adhesion during adhesive and deadhesive growth of LLC and LLC/R9 tumor cells were determined by flow cytofluorimetry. Novelty: For the first time, in vitro studies of tumor cells of the same origin, but with different metastatic potential, were carried out. Their comparative analysis made it possible to determine biophysical characteristics that can be considered as prognostic markers of the risk of metastasis.
