Laboratory prototypes of these DNA sensors were created, demonstrating detection limits for target DNA ranging from 0.001 to 0.1 nM, depending on the analysis variant. The linear dynamic range of the developed sensor devices spans from 0.001 to 10 nM. Approaches to improving the selectivity of the developed hybridization DNA sensors and strategies for their repeated use have been proposed. It was demonstrated that performing the analysis at an elevated temperature—corresponding to the optimal hybridization temperature of the immobilized oligonucleotide probe with a fully complementary sequence—resulted in a lowered detection limit for complementary oligonucleotides down to 0.001 nM, as well as a significant enhancement in selectivity in the presence of potential interferents (partially complementary and non-complementary nucleic acid sequences) in the test samples.
