popup.stage_title

popup.description1
The aim of the project is the synthesis, study of structural, physicochemical and catalytic properties of new catalytically active nanomaterials, namely, artificial enzymes (nanozymes); creation on their basis of new diagnostic methods and study of biological action of nanocomposites on eukaryotic cells (in particular, yeast). The creation of new nanomaterials with catalytic properties will allow the development of new methods of highly sensitive quantitative detection of a number of practically important substances (glucose, lactic acid, alcohols) for clinical diagnosis, sports medicine and food technology.

A laboratory prototype of a potentiometric biosensor based on creatinine deiminase and pH-sensitive field-effect transistors was constructed and its main bioanalytical characteristics were investigated. Optimization of the main bioanalytical characteristics of the laboratory prototype of a potentiometric biosensor for creatinine analysis (detection limit, sensitivity, linearity, selectivity, stability, reproducibility, etc.) was worked out and the phenomenon of degradation and reliability of biosensors under different analysis and storage conditions was investigated.
A laboratory prototype of a potentiometric biosensor based on recombinant CDI and pH-sensitive field-effect transistors and a laboratory prototype of a conductometric biosensor were tested on model and clinical samples of biological fluids from healthy donors and patients with various nephrological diseases and the results were validated using traditional analysis methods.
For the first time, the kinetics of the responses of a potentiometric biosensor based on a differential pH-sensitive field-effect transistor (pH-FET) for the determination of creatinine using the enzyme creatinine deiminase were simulated. The obtained model results were compared with the experimental responses of the pH-FET of the creatinine biosensor in the range of substrate concentrations of 1-10 mM and their good agreement was shown. For the first time, the possibility of using electrochemical biosensors based on recombinant creatinine deiminase for the non-invasive analysis of creatinine in human sweat, saliva and urine and the validation of the obtained results using traditional analysis methods were studied.
