Unique derivative cell lines of U-87 MG and U-251 MG with knockout or overexpression of specific CHI3L1 transcript isoforms were generated and comprehensively characterized for the first time. Distinct changes were identified in the expression of genes related to stemness, EMT, extracellular matrix remodeling, and therapy resistance. Differential angiogenic activity of the isoforms, their effects on neural cells, and secretion patterns under various culture conditions were demonstrated. Approaches for generating 3D spheroids and mixed cultures with neural cells and macrophages were optimized, and key features of their interactions were defined. The obtained results provide a scientific basis for developing more representative glioblastoma model systems and for further studies on the interactions between tumor, neuronal, and immune components of the microenvironment.
