popup.description1
The aim of the proposed project is to determine the functional role of the chitinase-like protein
CHI3L1 isoforms in the development and progression of glioblastomas, the impact on the main cellular components of the microenvironment (macrophages and neurons) using innovative 3D cell model systems.
popup.description2
At this intermediate stage of the project, several important scientific results were obtained, significantly advancing our understanding of the role of CHI3L1 isoforms in glioblastoma biology. Unique derivative cell lines of U-87 MG and U-251 MG with knockout or overexpression of specific CHI3L1 transcript isoforms were generated and comprehensively characterized for the first time. Distinct changes were identified in the expression of genes related to stemness, EMT, extracellular matrix remodeling, and therapy resistance. Differential angiogenic activity of the isoforms, their effects on neural cells, and secretion patterns under various culture conditions were demonstrated. Approaches for generating 3D spheroids and mixed cultures with neural cells and macrophages were optimized, and key features of their interactions were defined. The obtained results provide a scientific basis for developing more representative glioblastoma model systems and for further studies on the interactions between tumor, neuronal, and immune components of the microenvironment.
