popup.stage_title

popup.description1
Determination and characterization of features of functional activity and molecular mechanisms of its regulation of known isoforms of ribosomal protein kinase S6K1 - p55, p60, p70, p85 in normal and malignant transformation of cells.
popup.description2
he object of the study is ribosomal protein kinase S6 isoforms.
The purpose of the work is to determine the features of the functional activity of ribosomal protein kinase S6 isoforms -S6K1 in normal and pathological conditions.
Research methods - CRISPR/Cas9 genome editing system and shRNA interference, gene expression analysis by real-time PCR, Western blot analysis of protein expression and their phosphorylation, immunocytochemistry and confocal microscopy.
Polyclonal antibodies to ER were created and characterized. The effect of PI3K, mTOR, MAP inhibitors on the level of phosphorylation of the corresponding S6K1 substrates was studied. The effect of the expression of different isoforms was analyzed. S6K1 on the activity and level of expression of ER and, accordingly, the expression of ER-dependent genes. The effect of the expression of S6K1 isoforms on the efficiency of translation in cells with differential expression of S6K1 isoforms was studied. The translation efficiency was analyzed by determining the protein content and total and mRNA enriched in P bodies and stress granules of endogenous, as well as expressed full-length constructs of S6 kinases on model cell lines.
