Morphological and molecular verification of the samples was performed according to the current WHO classification, allowing the identification of IDH1 wild-type glioblastoma samples for further analysis. Protocols for tissue and cell processing and cryopreservation were optimized, ensuring high post-thaw viability and long-term storage of samples in the project biobank. A preliminary analysis of the immune cell composition in peripheral blood and tumor tissue revealed clear differences between systemic and local immune environments in glioblastoma. An important achievement of this stage was the establishment of glioblastoma organoid models derived from cryopreserved tumor tissue. These organoids retained key morphological and cellular markers of the original tumors, demonstrating their relevance as experimental models. Overall, the obtained results provide a solid experimental and resource platform for future studies of immune niches in glioblastoma and for the development of novel prognostic and therapeutic approaches.
