popup.description1
To obtaine an experimental series of polyhydroxyalkanoates produced by strains of bacteria of the Bacillaceae family for the
creation of composites with calcium phosphate biomaterials.
popup.description2
Optimized cultivation conditions for the strain Priestia endophytica CHAES 5_14 were established for the production of polyhydroxyalkanoates (PHAs). The synthesized PHA was identified as poly-β-hydroxybutyrate (PHB) with an average molecular weight of 181 kDa and thermal characteristics including a glass transition temperature of 2.3 °C, a crystallization temperature of 74.6 °C, melting temperatures of 155 and 164.5 °C, and a degradation temperature of 271 °C. A composite with hydroxyapatite (HA) was obtained using the solvent-casting method. Its mechanical properties were almost twofold lower than those of neat PHB: tensile strength of 14.3 ± 2 MPa with an elongation at break of 8.5 ± 1.2%, compared to 21.2 ± 3 MPa and 12.5 ± 4%, respectively. The biocompatibility of PHB and PHB/HA was demonstrated in vitro using the MA-104 cell line and in vivo in Wistar rats. Approaches for the fabrication of biocompatible composites with calcium phosphate materials (CPMs) for medical applications were developed. It was shown that pressing CPMs under a press onto a PHB film pre-wetted with chloroform yielded the best results for the formation of PHB/CPM composites. However, the non-uniform distribution of CPM particles over the PHB film surface indicates the need for further development of CPM deposition methods.
